I would like to load a 45 colt with a hunter 275 bullet and would like to know if anyone has any suggestion.

21-22 grs of H110/W296 with a mag pistol primer should work. I've shot that with a 300 gr cast in my Ruger Blackhawk.

I'm not familar with that bullet but I have loaded up and shot some Ranch Dog 290 gr with Lil'Gun and found it very accurate as well as pretty much capable of stopping a pig or deer. I see he used several powders in his development which included not only the Lil'Gun but Accurate 1680, Winchester 296, Norma R123, and Hodgdon H110. Those would also probably work in your case also.

I'm thinking you may need a slower burning powder and standard primers to take advantage of that much barrel. since I'm not really what I would consider much of a reloader, I'll defer to our pros.

You do need mag primers with H110/W296. They are the same powder just in a different container. With it you do not want to reduce your load below the minimum load as it may not function properly.
